Loop-seq is a high-throughput sequencing assay that measures DNA looping and can help explain how DNA bendability contributes to nucleosome organization.

Taekjip Ha’s group at Johns Hopkins University develops biophysical techniques such as super-resolution imaging for studying how single molecules behave in a biological system. Aakash Basu, a postdoctoral fellow in Ha’s lab, is particularly interested in DNA mechanics and how DNA sequence influences the mechanical properties of DNA and ultimately influences biological outcomes. Basu says, “The mechanical properties of DNA, such as bendability and torsional rigidity, can potentially determine how easy or how hard it is for critical macromolecules to interact with DNA.”
