Xiao et al.1 expressed concerns regarding the efficacy of gene delivery to the liver in a nonhuman primate (NHP) model of nonalcoholic steatohepatitis (NASH) via the use of adeno-associated virus (AAV) serotype 8 vectors in four papers we recently published in Nature Medicine2,3,4,5. We appreciate their concerns, but we also note that they are based on certain assumptions rather than a failure to validate our experimental results. In particular, the major concern of Xiao et al.1 is whether the pre-existence of neutralizing antibodies (NABs) against AAV8 was considered in our studies. Indeed, they were. And not only were AAV8 NAB titers measured in each and every single monkey used in our studies, but we also screened for a variety of infectious viruses and parasites as a standard practice before each monkey experiment. Specifically, only those monkeys negative for tuberculin, B virus, simian T cell leukemia virus type 1, SIV, simian retrovirus, endoparasites, ectoparasites, salmonella and shigella as well as having low or negative NAB titers were used in the papers in question2,3,4,5.
