Use of genome-wide CRISPR–Cas9 knockout screens in BRCA1/2-proficient prostate cancer cells enabled identification of previously unknown genes, loss of which affects PARP inhibitor response. MMS22L deletion caused PARP inhibitor hypersensitivity and CHEK2 conferred resistance.
