HIF-1 inhibition mitigates skin damage in CLE

Cutaneous lupus erythematosus (CLE) and lupus nephritis are both characterized by infiltrating myeloid cells, including T cells, that are implicated in tissue inflammation and damage. A new study has identified a pathogenic role for HIF-1 in regulating the cytotoxic activity of skin-infiltrating T cells in CLE.

Gene and protein expression analysis of lupus-prone MRL/lpr mice confirmed that skin-infiltrating CD4+ and CD8+ T cells had upregulated expression of HIF-1 and HIF-1-related genes. The skin-infiltrating T cells had a strong cytotoxic phenotype, including high expression of intracellular granzyme B, as well as a T helper 17 (TH17) phenotype.
