Calcium-triggered synaptic vesicle (SV) release adds membrane to the presynaptic active zone. An equal amount of membrane needs to be removed by endocytosis to maintain membrane homeostasis, but the mechanisms by which this occurs are poorly understood.

The SV calcium sensor synaptotagmin 1 (SYT1) interacts with exocytotic and endocytotic proteins as well as membrane lipids, and removal of SYT1 from synapses impairs both endocytosis and exocytosis, which suggests a potential involvement in the coupling of these two processes. Bolz et al. investigated the mechanisms of exocytosis–endocytosis coupling using genetic manipulation and optical imaging techniques in mouse hippocampal neurons.
