Anti-tumour antibody-producing plasma cells disseminate from tertiary lymphoid structures

The presence of intratumoural tertiary lymphoid structures (TLS) is associated with favourable prognosis in many cancers. Using spatial transcriptomic analysis, Meylan and colleagues identified intratumoural TLS as sites of in situ B cell maturation in renal cell carcinoma (RCC). They report that IgG and IgA-producing plasma cells disseminate from the TLS into the tumour tissue. Tumours containing TLS had high frequencies of IgG-producing plasma cells and apoptotic tumour cells with bound IgG antibodies, suggesting an anti-tumour effector mechanism. High numbers of IgG-labelled tumour cells were also associated with higher response rates and longer progression-free survival among patients with RCC who were treated with immune checkpoint inhibitors.
