Reversal of DNA replication forks stalled owing to replication stress is crucial for avoiding fork collapse and DNA breaks. Reversed forks are protected by dedicated factors to prevent their degradation, which results in genome instability and aggressive cancers. How fork stalling is sensed and reversal initiated is unclear. Dibitetto et al. show that DNA-dependent protein kinase catalytic subunit (DNA-PKcs) is essential for regulating fork dynamics in response to replication stress.
