MYC proteins are major drivers of tumorigenesis. Transcription is impeded by the formation of DNA supercoiling and associated topological barriers ahead of and behind RNA polymerases. Such torsional stress can be dissipated by DNA topoisomerases. Das et al. show that formation of a complex between MYC and the functionally complementing topoisomerases TOP1 and TOP2 promotes MYC-driven transcription.
