Actin branching maintains tissue-resident mast cells

Mast cells are tissue-resident myeloid cells that are important drivers of anaphylaxis, allergies, food avoidance and anti-parasite immunity. Although the developmental pathways of mast cells and their roles in immunity are widely accepted, the signals that maintain these cells in tissues remain insufficiently understood.

Inspired by previous observations that mast cells require integrin-mediated adhesion for their tissue positioning, Kaltenbach et al. hypothesize in this preprint (not peer reviewed) that cytoskeletal control mechanisms underlie the integrin-dependent organization of mast cells in tissues. The ARP2/3 complex, a heptameric multiprotein aggregate assembled around the ARPC4 protein, is crucial for cytoskeletal actin remodelling and branching, which inspired these authors to investigate whether ARPC4 and the ARP2/3 complex contribute to the tissue maintenance of mast cells.
