T cell priming and reactivation by dendritic cells (DCs) is an attractive therapeutic target for promoting or suppressing adaptive immune responses — for example, in cancer or autoimmune disease, respectively. Key mechanisms for the regulation of DCs in the hypoxic tumour microenvironment include the immunosuppressive function of the hypoxia-inducible factor HIF1α. A study in Nature by Sanmarco et al. now reports that lactate-induced HIF1α activation in the context of autoimmunity initiates a regulatory loop in DCs that limits immunopathology.
