Pancreatitis points to key role of dendritic cells in pancreatic cancer

Pancreatic ductal adenocarcinoma (PDAC) is highly resistant to immunotherapy, which has been ascribed to various factors, including low mutational burden and a hostile tumour microenvironment. PDAC is often associated with pancreatitis, but little is known about its contribution to cancer initiation or progression. In this preprint (not peer-reviewed), Mahadevan et al. show that pancreatitis alters the immune landscape of PDAC-bearing mice and suggest a crucial role for conventional type 1 dendritic cells (cDC1s) in promoting responses to immune checkpoint blockade (ICB).

The authors used syngeneic mouse models of orthotopic or genetically engineered PDAC with or without short-term or long-term pancreatitis (SP or LP, respectively). Proteomic analyses of pancreatic tissue from non-tumour-bearing mice with SP or LP showed an increase in infiltrating cDC1s, cDC2s and myeloid cells, and reduced numbers of T cells in pancreatic lymphoid structures. PDAC-bearing mice with pancreatitis also had an increase in infiltrating cDC1s and myeloid cells, which accelerated tumour initiation and progression.
