A pan-cancer signature for dysfunctional T cells

CD8+ tumour-infiltrating lymphocytes (TILs) are pivotal players in anti-tumour immunity but are often present in a dysfunctional state. Immune checkpoint blockade (ICB) therapies have revolutionized cancer treatment but only a fraction of patients respond, and biomarkers for effective ICB treatment are lacking. The tumour microenvironment shapes T cell functionality and thus the patient’s response to ICB. However, many of the factors that lead to dysfunctional TILs are unknown owing to heterogeneity between tumour types.
