Human DNA from a prehistoric artefact

Objects found in archaeological sites provide insights into human behaviour and culture from our evolutionary past. However, identifying the specific maker or user of ancient artefacts is currently not possible. In their study published in Nature, Essel et al. report a DNA extraction method that enabled the identification of an individual who handled a pendant approximately 20,000 years ago.

Current procedures for DNA extraction from ancient materials use harsh techniques that damage the integrity of precious samples. To resolve this issue, the authors screened several DNA extraction procedures for their ability to non-destructively isolate DNA from animal remains. They ultimately developed an elaborate extraction technique that resulted in minimal sample damage. Application of their protocol to ancient artefacts from existing collections resulted in the isolation of DNA of the animal they were made from. However, most objects were contaminated with DNA from modern humans, presumably introduced by handling during or after excavation.
