Numerous proteins controlling DNA damage and repair are known to influence cancer initiation, prognosis and therapeutic opportunities. Most studied proteins are tumour suppressors that counteract and repair DNA damage caused by intracellular processes and exogenous DNA-damaging carcinogens. A new study leverages bacterial genetic screens to identify endogenous proteins that instead promote DNA damage and mutagenesis. Functional extrapolations to human systems indicate diverse roles in human cancer mutagenesis, with potential clinical implications.
