Shining a light on age-related adrenal cancer progression

Adrenocortical carcinoma (ACC) is a rare aggressive cancer of the adrenal cortex with peak incidence in older adults and no effective targeted therapies. A study published in Nature Aging has used a mouse model of age-dependent adrenal cancer to identify a tumour-protective role for myeloid immune cells that is enhanced by androgens.

Ultrasound imaging was used to monitor adrenal size in Znrf3-cKO female mice. As expected, the adrenal glands increased in size by age 6 weeks, but after 6 weeks and up to age 12 weeks, the adrenal glands unexpectedly regressed in size. Measuring adrenal weight in male and female Znrf3-cKO mice at 4, 6, 9, 12 and 24 weeks of age confirmed these findings and suggested that Znrf3-deficient adrenal glands undergo a switch from hyperplasia to regression.
