Several chimeric antigen receptor (CAR) T cell constructs are approved for patients with B cell malignancies. These have the disadvantage of a reliance on autologous T cells derived from each patient, leading to prolonged manufacturing times, higher costs and a risk of manufacturing failure. Now, data from the phase I CALM trial provide early evidence of the safety and efficacy of UCART19, an allogeneic anti-CD19 CAR T cell construct featuring genetic ablation of TRAC and CD52, in patients with relapsed and/or refractory (R/R) B-cell acute lymphoblastic leukaemia (B-ALL).
