Castration-resistant prostate cancer (CRPC) arises when cancer cells can generate androgens intratumourally. Men with a germline missense variant of 3βHSD1, an enzyme that catalyses androgen precursor dehydroepiandrosterone (DHEA), develop CRPC with a shorter overall survival. Writing in The Journal of Clinical Investigation, Cui et al. uncover a role for cancer-associated fibroblasts (CAFs) in the induction of 3βHSD1 in prostate cancer cells.
