The ever-growing arsenal of single-cell sequencing technologies enables the comprehensive characterization of tumour cell states at molecular resolution. However, these methods require discontinuous measurements and therefore cannot closely monitor highly dynamic processes such as tumour cell invasion. Conversely, in vivo two-photon microscopy imaging can continuously study the dynamic behaviour of single tumour cells in their microenvironment, although without the depth of single-cell sequencing and with limitations related to photobleaching and phototoxicity.
