To examine the dynamics of intratumoural heterogeneity during tumour evolution, the authors extracted colorectal cancer tissue from multiple regions in three treatment-naive patients. Cell suspensions established from each tissue piece were then grown up in vitro as organoids. After 1 week in culture, each organoid was dissociated and sorted to obtain single cells from which clonal organoids were subsequently derived. For comparison, normal clonal organoids were also established at the same time using single crypt cells from adjacent healthy colorectal epithelium. Both normal and cancer-derived clonal organoids were then subjected to a number of analyses, including whole-genome and RNA sequencing.
