Antibody pre-adsorption can reduce the adverse effects of the biomolecular corona.

Developing nanotechnologies that improve the safety and efficacy of diagnostic and therapeutic approaches by targeting specific tissues and cells is a major goal in nanomedicine1. A common strategy is to enhance nanoparticle binding to cell receptors by coupling targeting moieties such as antibodies to their surface. Though this strategy has demonstrated promising outcomes in vitro, in vivo results have been less encouraging2. A recent multivariate analysis of the targeting efficacy of cancer nanotechnologies reported between 2006 and 2016 showed that only ~0.7% of the administered nanoparticles reached the solid tumour site1. Finding and overcoming the issues causing such a low in vivo efficacy would substantially accelerate the successful clinical translation of nanomedicines2.
