a Pre-immune and immune serum samples of individual mice were tested by immunoblotting analysis with blood-stage parasite lysate. Sera were diluted 1:200 and a representative example obtained with sera from a BALB/c mouse is shown. Blots derived from the same experiment and were processed in parallel. b Immune sera of mice were tested for parasite binding in IFA with in vitro cultured P. falciparum blood-stage parasites. As a representative example, results obtained with sera from a NMRI mouse are shown. The left panel shows differential interference contrast (DIC), the middle panel DNA staining with DAPI (blue) and the right picture is IgG immunofluorescence staining with Alexa Fluor 568 conjugated secondary antibodies (red). The parasite inhibitory anti-PfCyRPA mAb c1216 was used as positive control in both assays. Scale bar, 5 µm.
