IFN-λ and its receptor expression are hormone-dependent after intravaginal ZIKV infection. Six-week-old WT OVX mice were given hormone replacement regimens of vehicle, progesterone-only, or a combination of estradiol and progesterone. Mice were treated with 1 mg of anti-Ifnar1 mAb at day −1. On day 0, mice were inoculated via intravaginal route with 106 FFU of ZIKV (Dakar 41525); 1, 4, or 7 days after infection, total RNA was extracted, and mRNA expression analyzed by RT-qPCR (Ifnl2 ad; Ifnl3 eh; Il10rβ il; Ifnlr1 mp). Uninfected animals (for comparison) were harvested at the day 7 time point. Dotted lines indicate the LOD. Results are pooled from two or three experiments.
