TBT-dependent changes in DNA methylation might reflect changes in chromatin organization of F4 male gWAT. a Schematic representation of iso-Differentially Methylated Blocks (isoDMBs), and their association with regions with different base composition (%GC-isochores). isoDMBs represent chromosome domains punctuated by iso-directional Differentially Methylated Regions (DMR), i.e., whole genome consecutive, non-overlapping 100 bp windows showing significantly different MBD-seq read coverage between ancestrally TBT- and vehicle-exposed samples. isoDMBs aim to represent cases in which DMRs reflected regional changes in chromatin properties with a potential effect on the expression of genes therein. Hyper Browser was used to calculate the overlap between isoDMBs and each of the five groups defined with different GC content b or genes c, and to test their statistical significance using Monte Carlo tests58. The Jaccard index that measures the similarity between datasets was used to quantify the general overlap between isoDMBs and regions with different base composition or genes (see Methods). Expected Jaccard indexes were calculated after randomly rearranging isoDMBs location 10,000 times respecting isoDMBs, regions with different %GC, and gene length and chromosome assortment. p values represent the number of random simulations showing Jaccard indexes more extreme than observed ones.
