HCT116, HT29, SW480, and SW620 cells were cultured under sphere-forming conditions in the presence of 4 µM XMD8-92 or dimethyl sulfoxide vehicle control. a The mRNA levels of stemness-associated transcription factors were determined by qRT-PCR. b Steady-state protein levels of NANOG, OCT4, and SOX2 were confirmed by immunoblot analysis in HCT116-derived tumorspheres.
