The SmPill® manufacturing technique can ensure accurate and intact loading of vaccine formulation components. SmPill® minispheres were dissolved in PBS at 50 °C for 1 h. a SmPill® supernatants and CTB of known concentration were run on an SDS-polyacrylamide gel and stained with Coomassie blue. Bands corresponding to CTB can be found at 12 kDa and show that SmPill® can be accurately loaded with rCTB. b Following dissolution of SmPill® minispheres from two batches, GM1-specific ELISA was run in triplicate to confirm the binding capacity of rCTB inside the minispheres. CT in solution was shown as a technical positive control.
