CLASI-FISH could be a powerful technique for analysing the interactions of diverse bacterial species in complex communities

This new approach, combinatorial labelling and spectral imaging (CLASI)-FISH, is based on labelling a single bacterium with two or more probes, each of which is tagged with a distinct fluorophore. To test the approach, the authors synthesized eight versions of a FISH probe targeting Escherichia coli 16S rRNA, with a different fluorophore added to the 5′ end of each version. E. coli cells were separately labelled with all of the possible probe pairs, resulting in 28 combinations or 'label types'. Cells of each label type were then mixed and, following imaging by laser-scanning confocal microscopy, the authors were able to distinguish each of the 28 different label types within a single image.
