Paneth cells are packed together with LGR5 (Leu-rich repeat-containing G protein coupled receptor 5)-positive ISCs at the base of intestinal crypts and support stem cell function. Interestingly, calorie restriction was found to increase the numbers of Paneth cells and ISCs in mice. This observation, combined with the fact that the number of differentiated enterocytes was reduced following calorie restriction, indicated that reduced calorie intake promotes self-renewal but not differentiation of ISCs. In addition, ISCs from calorie-restricted mice displayed increased regenerative capacity, as assayed by the ability of isolated crypts to form organoid bodies (which are cell masses that contain all types of intestinal cells) in vitro.
