Nesprin-2 is one of the proteins of the LINC (linker of the nucleus and cytoskeleton) complex, which connects the cytoskeleton to the nuclear inner lamina and has a key role in force transmission and nuclear localization in migrating cells. The authors found that nesprin-2 binds directly to fascin in vitro and in different cell types, and that the localization of fascin at the NE depends on nesprin-2.

During cell migration, cell polarization involves the rearward displacement of actin cables that associate with nesprin-2, which positions the nucleus at the cell rear. The authors show that fascin–nesprin-2 binding, and thus the localization of fascin at the NE, are required for F-actin–nuclear coupling, nuclear polarization and nuclear movement.
