Detecting MoA by network dysregulation (DeMAND) uses gene expression profile (GEP) data to look at alterations to gene expression levels in response to treatment with a compound. For example, a compound that targets a transcription factor will also affect the expression of downstream targets of the transcription factor. Such changes can be assessed by calculating joint expression probabilities for one gene compared with another, and using additional mathematical approaches these changes can be assessed on a global scale. The genes that are most affected by a given compound or compound pairs can be ranked, and these predictions are verified by experimental data.
