Cyclic AMP and Growth of Fibroblasts: Effect of Environmental pH

Abstract

CULTURED normal fibroblasts grow logarithmically until a certain density is reached, at which time growth ceases. This cessation of growth has been termed contact or density dependent inhibition of growth1. Evidence suggests that cyclic AMP is important in the regulation of both phases of cell growth; rapidly growing cells have lower cyclic AMP levels than more slowly growing cells and the concentration of cyclic AMP rises at confluency in contact inhibited cells2–7. Accordingly, the addition of N6,O2′-dibutyryl cyclic AMP (a more potent analogue of cyclic AMP) slows the growth and decreases the saturation density5.
