Determination of the Nucleotide Sequence of a Fragment of Bacteriophage ϕX 174 DNA

Abstract

A general method for the direct sequencing of DNA fragments is described. Oligonucleotides produced by digestion with endonuclease IV are characterized by depurination and the sequencing completed by analysis of the products of partial digestion with exonuclease.
