Evidence that the Cohesive Ends of Mature λ DNA are Generated by the Gene A Product

Abstract

THE generation of the cohesive ends of λ DNA is generally believed to involve an enzyme or a group of enzymes which introduce two staggered single-chain scissions twelve bases apart (recent review in ref. 1). This hypothetical function has also been observed in the excision of a λ chromosome from a doubly lysogenic bacterium lacking all the known recombination functions, rec, red, and int2,3. This kind of excision cuts the prophage specifically between genes R and A, and is called Ter2.
