Promoter usage of BRCA1-IRIS

It was with great interest we read the recent article by ElShamy and Livingston1 describing BRCA1-IRIS, a novel product of the breast cancer susceptibility locus BRCA1. BRCA1-IRIS differs from previously characterized BRCA1 products owing to read-through into intron 11. This results in premature transcriptional termination and a different carboxy-terminal end of the encoded BRCA1-IRIS protein compared with the full-length BRCA1 p220 protein. ElShamy and Livingston1 report that the first 45 nucleotides of the BRCA1-IRIS transcribed sequence, upstream of exon 2, do not correspond to the known first exons (1a and 1b) of BRCA1. They claim that several homologues of these 45 nucleotides exist in the genome with one copy present about 24 megabases upstream of BRCA1 exons 1a and 1b. On the basis of these observations, the authors define a new first exon (termed exon 1c) and suggest that a separate promoter may govern the transcription of BRCA1-IRIS.
