Pacific Biosciences' PacBio technology was among the first single-molecule sequencing strategies to reach the market. The single-molecule real-time (SMRT) technology uses arrays of tiny 'zero-mode waveguides' to detect fluorescently labeled nucleotides in real time. The PacBio RS instrument created a sensation when it premiered at AGBT in 2010. Unfortunately, it was large and expensive, and although it consistently delivered longer reads than rival platforms, the throughput was low and error rates were an order of magnitude higher than with other instruments. As a result, short-read platforms from Illumina or Life Technologies of Carlsbad, California, have gained market share.
