Elucidation of each step in the pathway leading to Alzheimer's disease yields a protein that becomes a potential candidate for pharmacological intervention. Thus far, no compound has been developed that significantly affects the course of Alzheimer's, but it is axiomatic to researchers in the field that antagonists will be found that can modulate the activity of one or more of the proteins involved, and that this will “cure” the disease by slowing down the accumulation of toxic amyloid material. Recent papers identifying the β-secretase involved in amyloid precursor processing advance our understanding of the pathogenesis of Alzheimer's1,2,3,4, but the use of the new enzyme as a target for drug development will be challenging.
