A non-covalent inhibitor with pan-KRAS potential

The GTPase KRAS is one of the most commonly mutated proteins in cancer. Once considered undruggable, G12C variants of KRAS can now be targeted with two approved covalent inhibitors. However, these drugs are not effective against the many other variants that account for 80% of KRAS mutants. Now, a study in Nature reports the discovery of a small-molecule non-covalent KRAS inhibitor that reduced proliferation in cell lines and mouse models bearing a wide range of KRAS mutants.
