Ribosome inhibitor combats bacterial drug resistance

Several classes of antibiotics target the bacterial ribosome to block protein synthesis. However, ribosome modifications that disrupt antibiotic binding lead to resistance. Reporting in Cell, Zhang et al. have designed a series of macrolides that target the Mycobacterium tuberculosis (Mtb) ribosome and overcome a key resistance mechanism. The lead compound is efficacious in mouse models of tuberculosis (TB), both as a single agent and in drug combinations.
