Fighting ageing with immune checkpoint blockade

The accumulation of senescent cells in various tissues promotes inflammation and has been implicated in a variety of age-related diseases. However, little is known about the mechanisms underlying this accumulation and whether it can be therapeutically targeted. Now, writing in Nature, Wang et al. report that senescent cells expressing programmed cell death-ligand 1 (PDL1) are able to escape immune surveillance, resulting in their accumulation and associated inflammation. Moreover, targeting the senescent cell population using anti-programmed cell death protein 1 (PD1) therapy improved age-related conditions and ameliorated nonalcoholic steatohepatitis (NASH) pathogenesis in mice.
