Targeting cancers with chromosome instability

Cancer cells frequently undergo sustained errors in chromosome segregation during mitotic divisions. This results in a dynamic state of chromosomal instability (CIN), which drives tumour progression and is associated with metastasis, drug resistance and poor prognosis. Hong et al. now report in Nature that cancer cells with CIN are dependent on an inflammatory signalling pathway mediated by cyclic GMP–AMP synthase (cGAS) and its downstream effector, stimulator of interferon genes (STING). This dependency can be therapeutically targeted to prevent tumour growth in mouse models.
