Eliminating vaccine cross-reactivity

Dengue virus (DENV) and Zika virus (ZIKV) infection can induce cross-reactive antibodies, which recognize the immunodominant fusion loop epitope on envelope (E) protein and can promote antibody-dependent enhancement (ADE) of infection, a key concern in vaccine development. To circumvent this, Slon-Campos et al. designed stabilized triple-disulfide-bonded ZIKV E dimers, which do not expose the immunodominant fusion loop epitope. In mice, immunization with the ZIKV E dimers induced dimer-specific antibodies that protected against lethal ZIKV challenge. Importantly, mouse ZIKV E-dimer-immune serum did not exhibit cross-neutralization of DENV and did not induce ADE.
