Mlkl deficiency does not affect the formation of necrosome, but delays TNF+SmacM+zVAD-induced mitochondria fission. (A) Wild-type and Mlkl−/− MEFs were treated with TNF+SmacM+zVAD for different periods of time, lysed and immunoprecipitated with anti-Rip3 antibody. The immunoprecipitates and total cell lysates (TCL) were immunoblotted with antibodies as indicated. (B) Wild-type and Mlkl−/− MEFs or wild-type and Rip3−/− MEFs were treated with TNF+SmacM+zVAD for different periods of time. The cells were stained with Mitotracker Red and cell imaging was recorded.
