Stimulation of DNA synthesis by tumour promoter and pure mitogenic factors

Abstract

12-O-TETRADECANOYL PHORBOL-13-ACETATE (TPA), a potent tumour-promoting agent, stimulates the proliferation of various cell types, including fibroblasts, in cell culture (see ref. 1 for review). Recent studies have demonstrated that there is a close correlation between the potency of phorbol esters for promoting skin tumours and their ability to stimulate DNA synthesis in cultured cells2. The growth-stimulating activity of TPA is completely dependent on the presence of serum in the nutrient medium. This requirement for serum has been demonstrated in cultures of mouse epidermal cells3 and fibroblastic cells4,5. The role of serum in the cellular response to TPA remains unresolved. We report here that serum can be replaced by polypeptide hormones like epidermal growth factor (EGF)6, and insulin and by the growth factor isolated from the conditioned medium of SV40 BHK cells (fibroblast-derived growth factor, FDGF)7. Low concentrations of TPA and the growth-factors stimulate DNA synthesis in quiescent cultures of mouse and human cells in completely serum-free medium.
