Density-dependent phosphorylation of a specific protein in cultured cells

Abstract

THE phosphorylation–dephosphorylation reactions that regulate the biological activities of many proteins are sensitive to changes in physiological conditions1. Hormones regulate the phosphorylation of both membrane-bound2 and cytosolic proteins3,4, while the phosphorylation of histone5 and non-histone chromosomal proteins6 changes during different phases of the cell cycle. The cellular density of mammalian cells grown in culture can affect biological properties such as cell growth7, locomotion8, transport9 and enzyme activity10,11. We have examined the phosphorylation of endogenous proteins in cultured mammalian cells at different cellular densities as a means of determining whether cell–cell interactions might affect protein phosphorylation. We report here that the phosphorylability of a specific protein present in several diverse cell lines exhibits density-dependence.
