Subcellular Localization of 5-Hydroxytryptamine and Histamine in Blood Platelets

Abstract

BLOOD platelets of various species contain particular, dense, osmiophilic organelles (dense bodies) which are somewhat smaller (500 to 2200 Å) than the α-granules. By combining electron microscopy and 5-hydroxytryptamine determinations, including experiments with reserpine, we have provided strong evidence that the osmiophility of the organelles is caused by an accumulation of 5-hydroxytryptamine1–4. Experiments with tyramine confirmed this view5. Here, we report attempts to isolate the dense osmiophilic bodies of platelets by density gradient centrifugation. Furthermore, the distribution of 5-hydroxytryptamine and histamine in various subcellular platelet fractions, including the dense bodies, was determined.
