Abstract
EMBRYO apices of the spring wheat cv. ‘Koga II’ were used in the investigation described here. The shoot apical meristem together with its three leaf primordia was excised under aseptic conditions, but in some experiments the coleoptile was also removed with the apex. The excised apices were inoculated on to the surface of 5 ml. of a semi-liquid basal medium containing the mineral salts of Baldev1, 2 per cent sucrose and 0.3 per cent Difco ‘Noble’ agar in 3 in. × 1 in. soda-glass tubes and grown in the dark at 25° ± 1° C.
