Tissue Cultures on Solid Media

Abstract

WHEN tissue cultures of mouse tumours were routinely tested for mycoplasma by inoculation in Petri dishes containing Difco PPLO agar (7 parts), 25 per cent yeast extract (1 part) and horse serum (2 parts), a nodule of tumour cells 2 mm in diameter was seen on the agar surface of one dish examined after 14 days at 37° C. Several cell lines were then inoculated into this medium with substitution of foetal calf serum for horse serum and the addition of 100 µg/ml. each of penicillin and streptomycin.
