A Technique for obtaining Clone Cultures of Soil Amœbæ in Sterile Liquid Medium

Abstract

ON previous occasions free-living and parasitic amœbæ have been obtained free from their associated bacteria by sterilization of the cysts either by washing in bactericidal chemicals1,2 or in antibiotics3,4. The technique I have used for isolating clones of soil amœbæ and growing them in sterile peptone solution involves the treatment of the trophozoite stage with antibiotics, making use of the fact that the amœbæ are more tolerant to many antibiotics than are certain bacteria.
