Highly Active Triphenylethylene Derivatives

Abstract

Triphenylethylene1 and several active derivatives2–4 have been shown to possess a prolonged œstrogenic activity provided that a dose well above the threshold is administered. Among these, αα-di-(p-ethoxyphenyl)-β-phenylbromoethylene4 has been especially noted for its considerable prolonged œstrogenic activity when administered orally. A considerable prolonged action has also been noted5 when a dose well above the threshold of diethylstilbœstrol dimethyl ether is administered subcutaneously. Emmens6 has attributed the prolonged action shown on injection of the higher doses of methyl ethers of diethylstilbœstrol and of œstrone to “the slowness with which the body effects hydrolysis of such ethers, without which they cannot be esterified and excreted”.
