Effect of DSG3-RNAi on tumor xenograft BALB-C/nu mice. (a) The mice were subcutaneously injected with 107 KB cancer cells. After 3 days, 50 μg of either DSG3-RNAi plasmid or the control vector plasmid in a total of 50 μl normal saline solution was injected intravenously, followed by a booster injection with 25 μg of plasmid twice a week for 6 weeks. The tumors were monitored for total of 60 days. The tumor volume was calculated as length × width × height, each dimension having been measured by calipers. Tumor volume was significantly greater at 56 days in the mice given control vector as compared with those given DSG3-RNAi (two-sided t-test, P<0.001). (b) Six weeks after grafting, tumors were removed and subjected to IHC analysis of DSG3 (radish color) and counterstained with HE (blue). Hair follicle tissues were also stained as control to ensure the successfulness of IHC staining.
