DSG3 expression was knockdown by RNAi. (a) Two plasmids clones of RNAi were designed: DSG3-RNAi-1 (clone 2323) and DSG3-RNAi (clone 2761), with their sense and antisense hairpin structures illustration. (b) Differential effectiveness of suppressing DSG3 expression in the two clones of DSG3-RNAi plasmids was shown in OECM1 cells for example. For each experiment, 5 × 105 cells were seeded in a 10-mm dish plate and transfected with two clones of RNAi or vector plasmids into OECM1 cells. Cells were further cultured for 1 day and the cellular proteins were extracted for the determination of DSG3 expression by Western blot analysis. (ce) Effects of DSG3-RNAi on DSG3 protein expression in OECM1, KB and BM1 cells, respectively. Cells were seeded with 5 × 105 cells, transfected with DSG-RNAi and further cultured for 4 days. Cellular proteins were extracted for the determination of DSG3 expression by Western blot analysis.
